Changes of Serum Protein Composition in Rats After Portacaval Shunt

Abstract
Surgical treatment of esophageal varix bleeding by portacaval shunt has been widely practised. The effect of deprivation of portal blood flow from the liver has been extensively studied. Since the time of Eck, investigators have been intrigued by the physiologic changes induced by portacaval; shunt, in which the liver is partially devascularized by diversion of the splanchnic-change-drainage directly into the systemic venous system. Although there is general agreement regarding the pathologic modifications of liver function of following producing Eck fistula, many authors have reported the changes of serum protein composition after portacaval shunt differently.
With remarkable advancement of microvascular surgical technique and our experimental experience of portacaval transposition in rat, construction of author¢¥s experimental model became to be feasible.
Author performed this experimental work to investigate the changes of serum protein composition, especially on gamma globulin by paper electrophoresis in the experimental models of portacaval shunt in Korean hybrid rats.
All the experimental rats: were divided into 3 groups; control, the 1 week and the 2 weeks group after portacaval shunt.
The results were as follows;
1. The serum protein level was albumin; 3.2¡¾0.4(g/dl), globulin; 5.5¡¾0.7(g/dl), total protein-, 8.7¡¾0.5 (g/dl) in control group, albumin; 2.9¡¾0.4 (g/dl), globulin; 6.0¡¾0.3(g/dl), total protein; 8.9¡¾0.4(g/dl) in the 1 week group after portacaval shunt, albumin; 2.5¡¾0.3 (g/dl), globulin; 6.1¡¾0.5(g/dl), total protein; 8.6¡¾0.3(g/dl) in the 2 weeks group after portacaval shunt (Table I).
2. The serum protein composition of control group was albumin; 34.6¡¾3.9%, a,-globulin; 3.9¡¾1.1%, a2-globulin; 8.7¡¾2.2%, p-globulin; 30.2-7.2%, 1¢¥-globulin; 11.1¡¾-1.3% (Mean¡¾ S.D.) (Table ff ).
3. The serum protein composition of the 1 week group after portacaval shunt was albumin; 32.9¡¾4.0%, a,-globulin; 21.0¡¾3.1%, a2-globulin; 7.4¡¾1.5%, j3-globulin;-28.7¡¾ 4.4%, 1¢¥-globulin; 10.5¡¾2.6% (Mean¡¾S.D.) (Table 1).
4. The serum protein composition of the 2 weeks group was albumin; 28,6¡¾13.7%,
a2-globulin; 18.0¡¾2.8%, a2-globulin; 8.3=1.8%, fl-globulin: 20.2_¡¾9.2%, 7-globulin; 22.2¡¾. 5.9%¢¥ (Mean S.D.) (Table AT).
With the above results, serum albumin level in rat with portacaval shunt was markedly decreased but total protein level unchanged. The a3-globulin and r-globulin were increased and a2-globulin level unchanged. The A-globulin was rather decreased.
